The cutaneous basement membrane is known to play an important role in epidermal differentiation, epidermal- matrix adhesion, the control of selective permeability and physical barrier. However, few studies have addressed the factors that influence the formation of cutaneous basement membrane. Vitamin C and E are known to influence on the epidermal proliferation and differentiation still, there have been no study about obvious relationship between vitamin C or E and formation of basement membrane. This laboratory study was designed to elucidate the effects of vitamin C and E on the formation of the cutaneous basement membrane. Keratinocytes were cultured on collagen matrix including fibroblasts in the presence or absence of vitamin C or E at the air-liquid interface. After 2 weeks of culture, immunohistochemical stainings for the components of the basement membrane and electron microscopic studies of the basement membrane zone were performed. In living skin equivalent treated with vitamin C, type IV collagen, integrin α6, integrin β4 and laminin 5 were expressed, however, type VII collagen was not expressed. These basement membrane components were more strongly expressed than in the absence of vitamin C. In addition, elements of the basement membrane zone such as lamina lucida, lamina densa and anchoring fibrils were formed. In living skin equivalent treated with vitamin E, type IV collagen, integrin α6, integrin β4 and laminin 5 were all expressed. However, these components were more weakly expressed than in the living skin equivalent treated with vitamin C. Lamina lucida, lamina densa and anchoring fibrils were also formed.
